Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.

Psychology Minds are constructed entities, which have general parameters fixed by their constructors (other Minds) before 'birth', not unlike biological beings. A wide variety of characteristics can be and are manipulated, such as introversion-extroversion, aggressiveness (for warships) or general disposition. However, the character of a Mind evolves as well, and Minds often change over the course of centuries, sometimes changing personality entirely. This is often followed by them becoming eccentric or at least somewhat odd. Others drift from the Culture-accepted ethical norms, and may even start influencing their own society in subtle ways, selfishly furthering their own views of how the Culture should act. Minds have also been known to commit suicide to escape punishment, or because of grief. Minds are constructed with a personality typical of the Culture's interests, i.e. full of curiosity, general benevolence (expressed in the 'good works' actions of the Culture, or in the protectiveness regarding sentient beings) and respect for the Culture's customs. Nonetheless, Minds have their own interests in addition to what their peers expect them to do for the Culture, and may develop fascinations or hobbies like other sentient beings do.
=== United States === In 1988, there was a confirmed death from CJD of a person from Manchester, New Hampshire. Massachusetts General Hospital believed the person acquired the disease from a surgical instrument at a podiatrist's office. In 2007, Michael Homer, former Vice President of Netscape, had been experiencing consistent memory problems, which led to his diagnosis. In August 2013 the British journalist Graham Usher died in New York of CJD. In September 2013, another person in Manchester was posthumously determined to have died of the disease. The person had undergone brain surgery at Catholic Medical Center three months before his death, and a surgical probe used in the procedure was subsequently reused in other operations. Public health officials identified thirteen people at three hospitals who may have been exposed to the disease through the contaminated probe but said the risk of anyone contracting CJD is "extremely low". In January 2015, former speaker of the Utah House of Representatives Rebecca D. Lockhart died of the disease within a few weeks of diagnosis. John Carroll, former editor of The Baltimore Sun and Los Angeles Times, died of CJD in Kentucky in June 2015, after having been diagnosed in January. American actress Barbara Tarbuck (General Hospital, American Horror Story) died of the disease on December 26, 2016. José Baselga, clinical oncologist having headed the AstraZeneca oncology division, died in Cerdanya, March 21, 2021, from CJD.
== Self-assembling peptides versus carbon nanotubes == Carbon nanotubes (CNTs) are another type of nanomaterial that have attracted much interest for their potential to serve as building blocks for bottom-up applications. They have excellent mechanical, electrical, and thermal properties and can be fabricated to a wide range of nanoscale diameters, making them attractive and appropriate for the development of electronic and mechanical devices. They demonstrate metal-like properties and can act as remarkable conductors. However, there are several areas where peptides have advantages over CNTs. One advantage is that peptides have almost limitless chemical functionality compared with the very limited chemical interactions that CNTs can perform due to their non-reactiveness. Furthermore, CNTs exhibits strong hydrophobicity which results in a tendency to clump in aqueous solutions and therefore have limited solubility; their electrical properties are also affected by humidity, and the presence of oxygen, N2O, and NH3. It is also difficult to produce CNTs with uniform properties and this poses serious drawbacks as the reproducibility of precise structural properties is a key concern for commercial purposes. Lastly, CNTs are expensive, with prices in the range of hundreds of dollars per gram, rendering most applications commercially unviable.
Many proteins are involved in the process of cell signaling and signal transduction. Some proteins, such as insulin, are extracellular proteins that transmit a signal from the cell in which they were synthesized to other cells in distant tissues. Others are membrane proteins that act as receptors whose main function is to bind a signaling molecule and induce a biochemical response in the cell. Many receptors have a binding site exposed on the cell surface and an effector domain within the cell, which may have enzymatic activity or may undergo a conformational change detected by other proteins within the cell. Antibodies are protein components of an adaptive immune system whose main function is to bind antigens, or foreign substances in the body, and target them for destruction. Antibodies can be secreted into the extracellular environment or anchored in the membranes of specialized B cells known as plasma cells. Whereas enzymes are limited in their binding affinity for their substrates by the necessity of conducting their reaction, antibodies have no such constraints. An antibody's binding affinity to its target is extraordinarily high. Many ligand transport proteins bind particular small biomolecules and transport them to other locations in the body of a multicellular organism. These proteins must have a high binding affinity when their ligand is present in high concentrations, and release the ligand when it is present at low concentrations in the target tissues.

Classical EDS is characterized by extremely elastic skin that is fragile and bruises easily, and hypermobility of the joints. Molluscoid pseudotumors (calcified hematomas that occur over pressure points) and spheroids (cysts that contain fat occurring over forearms and shins) are also often seen. A side complication of the hyperelasticity presented in many EDS cases makes wound closure on its own more difficult. Sometimes, motor development is delayed and hypotonia occurs. The variation causing this type of EDS is in the genes COL5A2, COL5A1, and less frequently COL1A1. It involves the skin more than hEDS. In classical EDS, large variation in symptom presentation is seen. Because of this variance, EDS has often been underdiagnosed. Without genetic testing, healthcare professionals may be able to provide a provisional diagnosis based on careful examination of the mouth, skin, and bones, as well as by neurological assessment. A good way to begin the diagnostic process is by reviewing a person's family history. EDS is an autosomal dominant condition, so it is often inherited from parents. Genetic testing remains the most reliable way to diagnose EDS. No cure for type 1 EDS has been found, but a course of non-weight-bearing exercise can help with muscular tension, which can help correct some EDS symptoms. Anti-inflammatory drugs and lifestyle changes can help with joint pain. Lifestyle choices should also be made with children who have EDS to try to prevent wounds to the skin. Protective garments can help with this.
Source Filmmaker (SFM) is a free 3D computer graphics software tool developed and published by Valve for creating animated films, which uses the Source game engine. Source Filmmaker has been used to create many community-based animated shorts for various Source games, such as Team Fortress 2, the Left 4 Dead series, and Half-Life 2.
== Side-effects == The most common side-effects are upper respiratory tract infection, diarrhea, combined edema/peripheral edema and headache, respectively. Most clinical adverse events were similar between groups treated with pioglitazone in combination with metformin and those treated with pioglitazone monotherapy.
=== North Korean involvement === In August 2018, Reuters reported that a confidential United Nations investigation had found the North Korean government had failed to discontinue its nuclear and missile delivery programs, and in conjunction, was "cooperating militarily with Syria" and was "trying to sell weapons to Yemen's Houthis". In August 2019, the South Korean National Intelligence Service had tracked the Scuds missiles (used to attack Saudi Arabia) back to North Korea. In January 2024, South Korea's Yonhap News Agency reported that North Korea had evidently shipped weapons to Houthis via Iran, based on the writings in Hangul script that were found on missiles launched towards Israel. North Korea considers the Houthis as a "resistance force". In March 2025, after the airstrikes by the US Air Force commenced, North Korean Ambassador to Egypt, Ma Dong-hee, who is also accredited to Yemen, condemned the attacks on the Houthis as a threat to regional and global order.

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
It is a synthetic peptide analog of human growth hormone-releasing hormone. It is used clinically to reduce excess visceral abdominal fat in adults with HIV-associated lipodystrophy. It works by stimulating pituitary growth hormone release.
