Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.

Identity and purity are assessed by reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass and detects chemical modifications. Peptide mapping and amino acid analysis can verify sequence integrity. Water content is measured by Karl Fischer titration, and residual solvents may be checked by gas chromatography. These methods together support batch-to-batch consistency and routine quality control.
Lyophilized tesamorelin is generally stored refrigerated at temperatures between 2 and 8 degrees Celsius. The solid form is comparatively stable when kept dry and protected from light. Moisture uptake can promote aggregation and degradation, so sealed containers with desiccant are common. Researchers typically avoid repeated temperature cycling, which may stress the peptide. Documentation accompanying reference materials usually specifies a shelf life under these conditions.
Receptor-level activity begins when the peptide binds the GHRH receptor, a class B G-protein-coupled receptor found on pituitary somatotroph cells. Occupancy triggers Gs-mediated activation of adenylyl cyclase and a rise in intracellular cyclic AMP, which in turn promotes synthesis and pulsatile release of growth hormone. Because the compound acts upstream of the growth hormone axis rather than supplying hormone directly, its effect depends on intact pituitary function. Binding studies in cell culture and animal models have established this pathway; the detailed kinetics of receptor recycling in humans remain less well characterized.
Physicochemical behavior is dominated by the peptide backbone. The molecule is hydrophilic and carries a net positive charge near neutral pH, owing to several arginine and lysine residues. In solution it adopts a largely unstructured conformation, and aggregation is a known concern for peptide products of this size. Oxidation of methionine and deamidation of asparagine or glutamine residues are the principal chemical degradation routes. These liabilities shape how the material is formulated, handled, and analyzed, and they explain why lyophilized presentations are common in research settings.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, built from 44 amino acids. Its sequence follows the natural human GHRH(1-44) backbone, with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification blocks recognition by dipeptidyl peptidase IV, the enzyme that rapidly truncates the native hormone in circulation. The result is a molecule with a substantially longer plasma residence time than unmodified GHRH, which makes it practical for clinical and laboratory study.
A way to provide an investigational therapy to a patient who is not eligible to receive that therapy in a clinical trial, but who has a serious or life-threatening illness for which other treatments are not available. Compassionate use trials allow patients to receive promising but not yet fully studied or approved therapies when no other treatment option exists. Also called expanded access trial. == Physiology == FVIII is a glycoprotein procofactor. Although the primary site of release in humans is ambiguous, it is synthesized and released into the bloodstream by the vascular, glomerular, and tubular endothelium, and the sinusoidal cells of the liver. Hemophilia A has been corrected by liver transplantation. Transplanting hepatocytes was ineffective, but liver endothelial cells were effective. In the blood, it mainly circulates in a stable noncovalent complex with von Willebrand factor. Upon activation by thrombin (factor IIa), it dissociates from the complex to interact with factor IXa in the coagulation cascade. It is a cofactor to factor IXa in the activation of factor X, which, in turn, with its cofactor factor Va, activates more thrombin. Thrombin cleaves fibrinogen into fibrin which polymerizes and crosslinks (using factor XIII) into a blood clot. The factor VIII protein has a half-life of 12 hours in the blood stream when stabilized by the von Willebrand factor. No longer protected by vWF, activated FVIII is proteolytically inactivated in the process (most prominently by activated protein C and factor IXa) and quickly cleared from the blood stream. Factor VIII is not affected by liver disease. In fact, levels usually are elevated in such instances.
Ion exchangers can have binding preferences for certain ions or classes of ions, depending on the physical properties and chemical structure of both the ion exchanger and ion. This can be dependent on the size, charge, or structure of the ions. Common examples of ions that can bind to ion exchangers are:
=== Termination === The phosphorylation of PAR1's cytoplasmic tail and subsequent binding to arrestin uncouples the protein from G protein signaling. These phosphorylated PAR1s are transported back into the cell via endosomes where they are sent to Golgi bodies. The cleaved PAR1s are then sorted and transported to lysosomes where they are degraded. This internalization and degradation process is necessary for the termination of receptor signaling.

Mummies have been discovered in Korea that have been dated to around the 15th to 19th centuries, during the Joseon period. This is thought to be because of the rise of Neo-Confucianism in Korea, which prescribed certain burial practices that created conditions favorable to mummification. However, this mummification was likely unintentional, and even culturally seen as ominous and undesirable. Mummification likely occurred due to a number of factors. One such factor was the airtight seal (especially as the Korean climate is unfavorable to mummification) around the bodies, which was achieved using a mix of lime, clay, and sand. Recreations of the sealing process found that chemical reactions with the lime possibly caused a high and prolonged release of heat, which killed bacteria in the bodies. In addition, a large amount of clothing was usually placed inside tombs, which led to a shortage of oxygen inside. Mummies buried using the lime mixture sealing technique reportedly have especially excellently preserved soft skin and hair, which has enabled medical and genetic studies to be performed. Insights have been offered into the lifestyles and pathologies of Korean people during this period. Specific diseases for each individual have been identified.
=== Diet and meal timing === A Mediterranean-style eating pattern emphasizing vegetables, fruits, whole grains, legumes, nuts, and unsaturated fats—is associated with improvements in blood pressure, lipids, insulin sensitivity, and cardiovascular risk. Reduced-carbohydrate approaches may lower glucose and promote weight loss in insulin-resistant individuals. Evidence on meal timing suggests time-restricted eating or avoidance of late-night meals can modestly improve glycaemic and lipid markers, though long-term data are limited. Guidance recommends tailoring dietary advice to personal preference, culture, and access to improve adherence.
Progesterone levels in the blood were found to be lower in those who had higher weight and higher BMI among those who became pregnant through in vitro fertilization. Current data shows that micronized progesterone, which is chemically identical to the progesterone produced in the human body, in combination with estrogen in menopausal hormone therapy does not seem to have significant effects on venous thromboembolism (blood clots in veins) and ischemic stroke (lack of blood flow to the brain due to blockage of a blood vessel that supplies the brain). However, more studies need to be conducted to see whether or not micronized progesterone alone or in combined menopausal hormone therapy changes the risk of myocardial infarctions (heart attacks). There have not been any studies done yet on the effects of micronized progesterone on hair loss due to menopause. Despite suggestions for using hormone therapy to prevent loss of muscle mass in post-menopausal individuals (age 50+), menopausal hormone therapy involving either estrogen alone, or estrogen and progesterone combined, has not been found to preserve muscle mass. Menopausal hormone therapy also does not result in body weight reduction, BMI reduction, or change in glucose metabolism.
Thermal stability of proteins has traditionally been investigated using biochemical assays, circular dichroism, or differential scanning calorimetry. Biochemical assays require a catalytic activity of the protein in question as well as a specific assay. Circular dichroism and differential scanning calorimetry both consume large amounts of protein and are low-throughput methods. The Thermofluor assay was the first high-throughput thermal shift assay and its utility and limitations has spurred the invention of a plethora of alternate methods. Each method has its strengths and weaknesses but they all struggle with intrinsically disordered proteins without any clearly defined tertiary structure as the essence of a thermal shift assay is measuring the temperature at which a protein goes from well-defined structure to disorder.
=== Early work === Thomson's prize-winning master's work, Treatise on the motion of vortex rings, shows his early interest in atomic structure. In it, Thomson mathematically described the motions of Lord Kelvin's vortex theory of the atom. Thomson published a number of papers addressing both mathematical and experimental issues of electromagnetism. He examined the electromagnetic theory of light of James Clerk Maxwell, introduced the concept of electromagnetic mass of a charged particle, and demonstrated that a moving charged body would apparently increase in mass. Much of his work in mathematical modelling of chemical processes can be thought of as early computational chemistry. In further work, published in book form as Applications of dynamics to physics and chemistry (1888), Thomson addressed the transformation of energy in mathematical and theoretical terms, suggesting that all energy might be kinetic. His next book, Notes on recent researches in electricity and magnetism (1893), built upon Maxwell's Treatise upon electricity and magnetism, and was sometimes referred to as "the third volume of Maxwell." In it, Thomson emphasized physical methods and experimentation and included extensive figures and diagrams of apparatus, including a number for the passage of electricity through gases. His third book, Elements of the mathematical theory of electricity and magnetism (1895) was a readable introduction to a wide variety of subjects, and achieved considerable popularity as a textbook.

The Cold War had provided external stabilizing pressures. Both the United States and the Soviet Union had a vested interest in Yugoslavia's stability, ensuring it remained a buffer state in the east–west divide. This resulted in financial and political support for its regime. When the Cold War ended, this external support evaporated, leaving Yugoslavia more vulnerable to internal divisions. As Yugoslavia fragmented, the wars began after Slovenia and Croatia declared independence in 1991. Serbia, under Slobodan Milošević, opposed these moves. The Bosnian War (1992–1995) was the most brutal of the Yugoslav Wars, characterized by ethnic cleansing and genocide. International organizations, including the United Nations, struggled to manage the violence. NATO eventually intervened with airstrikes in Bosnia (1995) as part of Operation Deliberate Force and later in Kosovo (1999) as part of Operation Allied Force. These interventions marked the transition of NATO as a deterrent to the Soviet Union, to also functioning at the time as an active peacekeeping and conflict-resolution force.
August 15, 2009: Argentina Even though official government sources state that Argentina's GDP will actually grow this year (and have only recognized a GDP shrinking in June of 0.4% and in July, of 0.3%; both compared to last year's same months), private consulting firms state that Argentine economy has actually been in recession since October 2008. According to them, Argentine GDP shrank 2.9% in the last quarter of 2008, 1.6% in the first quarter of 2009, and 0.6% in the second one. It's widely known in Argentina that the official statistics office in charge of producing this information, INDEC, is not very reliable for trustworthy statistical information, due to the government's mismanagement of the office.
This was achieved in 2010 when mature cardiac fibroblasts were reprogrammed directly into cardiomyocyte-like cells. This was done using three transcription factors: GATA4, Mef2c, and Tbx5. Cardiac fibroblasts make up more than half of all heart cells and are usually not able to conduct contractions (are not cardiogenic), but those reprogrammed were able to contract spontaneously. The significance is that fibroblasts from the damaged heart or from elsewhere, may be a source of functional cardiomyocytes for regeneration. Simply injecting functioning cardiac cells into a damaged heart is only partially effective. In order to achieve more reliable results, structures composed of the cells need to be produced and then transplanted. Masumoto and his team designed a method of producing sheets of cardiomyocytes and vascular cells from human iPSCs. These sheets were then transplanted onto infarcted hearts of rats, leading to significantly improved cardiac function. These sheets were still found to be present four weeks later. Research has also been conducted into the engineering of heart valves. Tissue-engineered heart valves derived from human cells have been created in vitro and transplanted into a non-human primate model. These showed a promising amount of cellular repopulation even after eight weeks, and succeeded in outperforming currently-used non-biological valves. In 2021, researchers demonstrated a switchable iPSCs-reprogramming-based approach for regeneration of damaged heart without tumor-formation in mice.
== Function == Nicotinamide phosphoribosyltransferase (iNAMPT) catalyzes the condensation of phosphoribosyl pyrophosphate with nicotinamide (NAM) to yield nicotinamide mononucleotide (NMN) and pyrophosphate (PPi). This is the first step in the biosynthesis of nicotinamide adenine dinucleotide (NAD+). This reaction allows a salvage pathway, in which NAM is recycled from enzymes that use NAD+ (sirtuins, PARPs, CD38) as these produce NAM as a waste product. Thus this reaction is a major source of NAD+ production in the body. De novo synthesis of NAD+ from tryptophan occurs only in the liver and kidney, overwhelmingly in the liver.
The government became increasingly authoritarian, and resistance movements, encouraged by Ethiopia, sprang up across the country, eventually leading to the Somali Civil War. Among the militia groups were the Somali Salvation Democratic Front (SSDF), United Somali Congress (USC), Somali National Movement (SNM) and the Somali Patriotic Movement (SPM), together with the non-violent political oppositions of the Somali Democratic Movement (SDM), the Somali Democratic Alliance (SDA) and the Somali Manifesto Group (SMG).

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
